Phenotypic screening assays are pivotal in drug discovery, identifying lead compounds based on their ability to induce changes in disease-relevant cells. This target-agnostic approach broadens the scope of “druggable” targets. Utilizing multi-parametric assays and live-cell imaging offers deeper insights into compound effects compared to traditional end-point methods.
Charnwood Discovery leverages these strategies with expertise in various cell types—including immortalized lines, iPSC-derived, and patient-derived cells—within both 2D and 3D models. Using live-cell imaging technologies like the Sartorius Incucyte® SX5, they study cellular functions such as migration, differentiation, and chemotaxis. Advanced imaging techniques and specialized algorithms enable tracking of processes like wound healing, DNA damage, and autophagy, generating rich data for candidate selection.
The poster highlights specific applications like scratch wound assays for assessing cell migration, chemotaxis assays to distinguish cytotoxicity effects, and high-content imaging to monitor DNA damage (e.g., γH2AX foci) and autophagy. Additionally, 3D culture systems for spheroid formation and analysis, underscoring the importance of physiologically relevant models in modern drug discovery.
